If you're talking about padlocked doors, they can only be opened from the side the padlock is on. From there, you can kick a few times, slice off the lock with your knife or just shoot it. If you're talking about the barricaded door where the blue medallions are, you can only unlock that later in the game. In fact, if a door is locked, you probably just need to progress a bit more until you find the key, or get around to the other side.
