For consistent results, yes. The knot will affect the way the pile leans, so if you go "upside down" on some and "right side up" on others (or over/under from another perspective) it will show in how the yarn lays with each other. It may look chaotic, messy, and like it has little "cowlick" or odd parts in the grain of the yarn.
