Megestrol acetate is a synthetic hormone used to treat certain types of cancer (most notably breast cancer) and certain types of anorexia (dysphagia) which can occur in people with AIDS. It is also used as a component of pet food as an appetite stimulant.

Mechanism of Action

Megestrol acetate is a progestin hormone. It works by binding to progesterone receptors in the cells, thus causing estrogenic and anti-estrogenic effects. It also stimulates the appetite and helps restore normal appetite.

How long does it take to work?

The effects of megestrol acetate usually occur within a few days, but may take up to a few weeks. The improvement in appetite may occur within one to two weeks.

Absorption

Megestrol acetate is rapidly and completely absorbed from the gastrointestinal tract after oral administration.

Dosage

Tablet:

Breast cancer: 160 mg/day
Endometrial carcinoma: 40-320 mg/day in divided doses.
At least 2 months of continuous treatment is considered an adequate period for determining the efficacy of Megestrol.

Oral Suspension: The recommended adult initial dosage of Megestrol Oral Suspension is 800 mg/day (20 ml/day).

Megestrol acetate is available in many forms with different doses, including 2.5, 5, 10, 20, 40, and 80 mg tablets, 10 mg/mL oral suspension, and 160mg/5mL suspension.

Administration
